The Desktop and Start menu Items are gone Server 2008 R2

Hello,  i am running a Server2008 R2 running the Roles of Active Directory Domain, Application server, dns server, file services, hyper-v, print and document services, remote desktop services and web server(IIS).  I am the only one that has access to the server and I have done no changes.  Yestarday an update to a program running on the system was sent to one of our employees.  The update is an exe file that is remained after putting it in the program folder.  I do not believe it caused the problem.  The Error was as follows:

C:\Windows\system32\config\systemprofile\Desktop refers to a location
that is unavailable. It could be on a hard drive on this computer, or on
a network. Check to make sure that the disk is properly inserted, or
that you are connected to the internet or your network, and then try
again.If it still cannot be located, the information might have been
moved to a different location

I have to say it is kinda feaking me out.
