Use a virtual/NAS drive as the c: drive to hold the operating system and programs on a server?

Is it possible to use a virtual drive as the c: drive to hold the operating system and programs on a server?

Our current C: drive only has 30gb and we need more space to support one of our programs which only uses the c: drive for temporary storage ( we are not allowed to change it). If we could use a virtual drive as the c: drive we could define as large as a  drive as we wanted.

Any comments are welcome
